The Nightmare Dungeon does have Treasure Chest rooms, just like the Undead Dungeon, but you'll have to consult a wiki to figure out which room it is, since there are several possible layouts depending on your spawn point.
Besides that, empirical evidence (from my personal experience) shows that certain characters, like Infernal Fiend, seem to spawn more often in the innermost parts of the dungeon (such as Treasure Chest rooms), while others (like the Void Incarnates and all their evolutions) will spawn fairly common everywhere (so you can just hang around the entrance and hit Explore).
I don't know if Overfiend Blade appears more often in the TC room or not, but he's a fairly rare find at the spawn point.
And of course, there's the Boss characters which only spawn in the Boss room.

Ok, updated my answer then xD Cus I searched for hours at the entrance and didn't encounter a single one. Different maps, perhaps.
