1= Heal player 1/Attack player 1
2= Heal player 2/Attack player 2
3= Heal player 3/Attack player 3
4= Heal player 4/Attack player 4
1v1:
Player 1= Player on the left
Player 2= Player on the right
2v2:
Player 1= Top left hand corner
Player 2= Top right hand corner
Player 3= Bottom left hand corner
Player 4= Bottom right hand corner
Jugg:
Player 1= Top left hand corner
Player 2= On the right
Player 3= Bottom left hand corner

You can set up your own custom hotkeys to the way you like it
I think hotkeys will be good because it is much quicker then using your mouse.
If you have lost your mouse/ broke you mouse or mousepad, you could use your keyboard instead of your mouse.
If you have a dodgy mouse and takes really long to move it, you could use your key board
