It affects both. As for calculation:

Player Stats + Initiative = P
Monster Stats + Initiative = M
First Strike Roll:
Player: X = Random # Roll (1,100) + P
Monster: Z = Random # Roll (1,100) + M
If X > Z, player goes first. If X = Z then there is a 50% chance of either player or monster going first. If Z >X, monster goes first. This means that if P exceeds M by 101 or more, the Player will always strike first.
So for a human explanation, the player and monster will roll random numbers between 1 to 100, and then whatever they rolled, P and M is added respectively. Whoever has the highest number has the right to go first. The most assured way to go first is to invest in multiple MAINSTATS and LUK, alongside using the ambush potion and an armor that provides initiative bonuses like Tempest Power Armor, Fortune Thief of Hippocampus Rider.
